CHAPTER [2] AMINO ACID AND PEPTIDES

**ysis of Amino Acid Mixture

Amino acids are usually separated on columns of cation-exchange resins in which the fixed anionic groups are first “charged” with Na+, an acid solution [PH 3.0] of the amino acid mixture is then applied to the column and allowed to percolate through at pH 3.0 the amino acids are largely cataions with net positive charge but they differ in the extent to which they are so ionized the cationic amino acids will therefore displace the bound Na+ ions from the resin particles at pH 3.0 the most basic amino acids [lysine, arginine, and the Ammonia] will displace Na+ first and will be bound to the resin most tightly and the most acids [glutamic acid, Serine] will be bound the least, the different amino acids will therefore move down the resin column at different rates the eluate fraction collected are **ysed quantitatively by means of the ninhydrin so that elution collection of fraction **ysis of each fraction and the recording of date are performed automatically in an amino acids **yzer,

Another method of separating amino acids is paper electrophoresis, in this process, a drop of solution of the amino acids mixture is placed on a filter-paper sheet, occurs which a high-voltage electrical field is applied, because of their different pK, value, the amino acids migrate in different directions and at different rates, depending on the pH of the system and the voltage applied, for example! At pH 1.0 Histidine, Arginine, and Lysine have charge of +2 moves more rapidly to the negatively charged cathode than all the other amino acids, which have a charge +1, At pH 6.0 the positively charges amino acids, all other will remain at origin since they are iso-electric at this pH

Histidine = C6H9N3O2 = 157.25 = 7.59 pH

Arginine = C6H14N4O2 = 177.5 = 10.74 pH

Lysine = C6H14N2O2 = 149.5 = 9.74 PH

For example, a1 = positive

            And b2 = negative

So = a1 / b2 = c1

Histidine = a1 = 157.25 and b2 = 46.25 Chromosomes

              = a1 / b2 = c1

              = 157.25 / 46.25 = C1

              = 157.25 / 2139.0625 = 0.07351351351351351351351351351351

              = 0.07 * +2 = 2.07

              = +2.0 moves more rapidly to the negatively charged cathode than all the other amino acids, by deeply function of Chromosomes,

For example, a2 = positive

            And (b2)2 = positive

So = a2 / (b2)2 = c2

Histidine = a2 = 157.25 and b2 = 46.25 Chromosomes

              = a2 / (b2)2 = c2

              = 157.25 / 46.25 = c2

              = 24727.5625 / 2139.0625 = 11.56

              = 11.56 - 7.59 pH = 3.97

              = 3.97 * +2 = 5.97

              = 6.0 = at pH 6.0 the positively charges amino acids, by deeply function of Chromosomes,

Actually the Chromosomes has very high language of functions, the Chromosomes are talk with each another, see to each another, and from this reason we can talk and see and other process of body,
