Amino Acid Sequence of Polypeptide Chain:
The next step in determination of the amino acid sequence is to fragment the intact polypeptide into smaller pieces; the method of choice is enzymatic hydrolysis using the proteolytic enzyme Trypsin, this enzyme catalyze the hydrolysis of only those peptide bonds in which the carboxyl group is donated by either a Lysine or Arginine residue,
Sue of overlapping from two different fragmentation procedures to establish the sequence of amino acids in a polypeptide chain of 21 residues,
Lysine = 149.5 m/g
Arginine = 177.5 m/g
Trypsin = 4(HCl) + CH2OH + H2O = 145 + 31.75 + 18.5 = 195.25
Peptide = C7H8N2O4 = 84 + 10 + 28 + 64 = 186 mole / 11.625 Carbon = 16 Peptide bonds
For example
= a2 / b2 = c2
= a2 = Trypsin and b2 = Peptide bonds = c2
= a2 = 195.25 and b2 = 16 = c2
= a195.25 / b16 = c2
= 38122.5625 / 256 = c2
= a / b = 148.91= c2
= a / b = 148.91 + 0.59 = 149.5
= a / b = 149.5 = Lysine
= a / b = 149.5 / 3.303 (H3) donated = 45.25
= 45.25 = COOH = carboxyl-group, the carboxyl group is donated by either a Lysine or Arginine residue

for example (Trypsin 195.25 / 90.75 a-amino acid = 2.15 * 10 cm = 21.5 residue),
the number of fragments products by trypsin cleavage can thus be predicted from the total number of Lysine or Arginine residues in the each chain, moreover all the fragments resulting from trypsin cleavage will have Lysine or Arginine residue at carboxyl-terminal position, the various fragments are the separated from each another by means of algebra, Each is then hydrolyzed and its amino acid content determined on another sample of each fragment
Now you should never forget that two chromosomes are jointed with each another into the cell for make pair of metabolism, if two chromosomes are feel presences of Carboxyl-terminal then the Carbon function will be activate into the one pair of chromosomes,

The amino terminal acid is then identified by reaction with 2,4 dinitrofluorobenzene or reagent by algebra, with the information the amino acid sequence of all the dipeptide and tripeptide fragments resulting from Trypsin cleavage will be known, the larger peptide from the Trypsin can then be sequenced by repeated algebra stop, in the manner of amino acid sequence of each of the fragments obtained by Trypsin cleavage of the chain can be establish,
