CHAPTER [3] PROTEINS

Proteins

(8/18/2016)

Charge and Chemical Properties of Amino acids and Proteins,

An understanding of protein requires knowledge of the ionizable side chain groups of the common amino acids, these ionizable groups common to proteins and amino acids, the acid form of the respective ionizable groups are on the left of the sum, while the respective properties base are on the right side, characteristic of the acid form is that nitrogen-containing groups are positively charged, whereas the acid forms that contain oxygen and sulfur atoms are natural,

Where acid group found                 Acid form = Base form,

COOH-terminal residue in peptide lysine = R-COOH = R-COO- + H+ = 3-5.5 pK

                                   Carboxyl Acid = Carboxylate

Left side = COOH = 45.25

Carboxylate = R-COO- + H+ = 57.25

  = a2 - b2 = c2

  = a2 (Left side) + b2 (Carboxylate) = c2

  = a2 (45.25) + b2 (57.25) = c2

  = a + b = 72.97 / 1.64 Optimum pH of stomach pepsin = 44.49

  = a + b = 44.49 - 0.44 () = 44

  = a + b = 44 = COO- = Carboxylate at right side

  = a + b = 44 = COO- + H+ = base form at right side

  = a + b = 44  + R = R- COO- + H+ = base form at right side

  = a + b = 3-5.5 pK = low acidic amino acids,

HUMAN CHROMOSOMES

Carboxylate at right side = R-COO- + H+ = 57.25

Human chromosomes = 46.25

  = a2 + b2 = c2

  = a2 (R-COO- + H+) + b2 (chromosomes) = c2

  = a2 (57.25) + b2 (46.25) = c2

  = a + b = (3277.5625) + b (2139.0625) = c2

  = a + b = 5416.625 = c2

  = a + b = 5416.625 / 73.59 = 73.60

  = a + b = 73.60 – 16.35 (O1) = 57.25

  = a + b = 57.25 = R-COO- + H+ = base form at right side,

  = a + b = 73.60 * 2.017 % negatively charged = 148.5 = Glutamine

  = a + b = 73.60 * 1.820 % negatively charged = 134 = Asparagine

  = a + b = 3-5.5 pK = low acidic amino acids,

The carboxyl acid-containing on side chain amino acids have relatively low pK value and are called the acidic amino acids, they are predominantly in their basic form and negatively charged at physiological pH,
