Proteins  

Some Chemical Reactions of the Amino acids, 

The amino group in amino acid react with “ninhydrin” on heating to give a blue compound, as reaction is stoichiometric, the amount of amino aicds present in a solution can be quantified by the high absorption of wavelength 570 nm, Proline an amino acid, also react with ninhydrin, but gives a yellow color with a maximum absorbance at 440 nm, 

 

PROLINE AND NINHYDRIN  

1 = Proline = (C5H9NO2), 117.25 m/g

2 = Ninhydrin = (C9H6O4), 179.5 m/g

 

  = a2 + b2 = c2 

  = a2 (Proline) + b2 (Amino acid) = c2 

  = a2 (117.25) + b2 (179.5) = c2 

 

  = a + b = (13747.5625) + b (32220.25) = c2 

  = a + b = 45967.8125 = c2 

  = a + b = 45967.8125 / 45.25 (COOH) = 1015.86 

  = a + b = 1016 + 84 (Amino acid) = 1100

  = a + b = 1100 / 2.5 (H2 hydrin) = 440 

  = a + b = 440 (nm) yellow color 

They give a yellow color with a maximum absorbance at 440 nm 

 

HUMAN CHROMOSOMES

PROLINE AND NINHYDRIN WITH CHROMOSOMES

  = a + b = 45967.8125 / 46.25 (Chromosomes) = 993.89

  = a + b = 1100 / 2.5 (H2 hydrin) = 440 

  = a + b = 440 (nm) yellow color 

They give a yellow color with a maximum absorbance at 440 nm

The fluorescamine reacts with amino acids in a 1:1 stochiometry at room temperature to give a fluorescent product the concentration of which can be determined with a spectra-flour-meter, the sensitivity of the fluoresce-amine assay is 10 to 100 times greater than ninhydrin assay can routinely measure 3 * 10-9 mol of an amino acid, the fluoresce-amine procedure can measure 10-10 to 10-11 mol an amino acid, 

 

A goodly number of specific chemical reaction can used to quantify specific amino acid side chain in a solution of free amino acid or in a denatured protein, the sakaguchi reaction (a-naphthol and sodium hypochlorite in alkaline solution) is used to color-metrically quantify the guanidine side chain in Arginine, Ehrlich reagent specifically gives a colorimetric assay for inside group of tryptophan, 

 

The Pauly regent gives a red color with the imidazole side chain of Histidine and the phenol side chain of Tyrosine,  

 

HISTIDINE TYROSINE AND NINHYDRIN  

1 = Histidine = C6H9N3O2 = 157.25 m/g

2 = Tyrosine = C9H11NO3 = 183.75 m/g

 

  = a2 + b2 = c2 

  = a2 (Histidine side chain) + b2 (Tyrosine side chain) = c2 

  = a2 (157.25)               + b2 (183.75) = c2 

 

  = a + b = (24727.5625) + b (33764.0625) = c2 

  = a + b = 58491.625 = c2 

  = a + b = 58491.625 / 45.25 (COOH) = 1292.63

  = a + b = 1293 / 2 (H2 hydrin) = 646.5

  = a + b = 646.5 (nm) red color 

The red color is equal to 647–700 (nm) wavelength

  = a + b = 58491.625 = c2 

  = a + b = 58491.625 / 46.25 (Chromosomes) = 1264.68

  = a + b = 1265 / 2 (H2 hydrin) = 632.5 

  = a + b = 632.5 (nm) red color 

The red color is equal to 647–700 (nm) wavelength
